Protein purification

Purification Process

Biortus has extensive experience in membrane proteins, soluble proteins, secreted proteins, inclusion body proteins, protein complexes, etc. It has successfully prepared over 6,000 proteins, covering all current hot drug targets. Biortus is equipped with more than 50 AKTA, offering diverse chromatographic purification methods such as affinity chromatography, ion exchange, hydrophobic and size exclusion chromatography. It also provides various QCs, including SDS-PAGE, Analytical SEC, LC-MS, SEC-MALS, and Mass photometry, enabling quick identification of a sample’s Mw, purity, homogeneity, and protein post-translational modifications. Below is the regular purification protocol for most proteins in Bioruts.

Biortus has multiple parallel laboratories, enabling purification of a large number of different protein targets simultaneously.

Automatic affinity chromatography not only enables high-throughput protein purification but also ensures the reproducibility across different batches.

With over 50 AKTA, diverse purification requirements can be met.

Quality Control

For quality control, SDS-PAGE was used for routine purity determination and NanoDrop was used for concentration measurement. We also employ methods such as Analytical SEC, SEC-MALS, and Mass Photometer to ensure the high quality of our protein products. Moreover, we can also perform buffer optimization or protein stabilization through ThermoFluor or NanoDSF upon request. Negative staining for cryo-electron microscopy can also be used for protein quality control. Besides conventional recombinant protein production, Biortus also has rich experience in customized protein modifications, such as Se-Met derivatization, isotope labeling, biotinylation, etc.
