Fluorescence Polarization Assay


Emission of fluorophore excited by linearly polarized light could remain polarized depending on its fluorescence lifetime and rotational correlation time. This phenomenon has led to the development of competition-based assays between fluorescently labelled ligands and test compounds for protein targets. The technique is frequently used in high-throughput screening and small molecule drug discovery as it is homogeneous and quantitative (allows calculation of IC50).
